STUDIES ON LEUKOCYTE ALKALINE PHOSPHATASE

Abstract
The authors carried out cytochemical studies for the demonstration of leukocyte alkaline phosphatase using a new substrate, naphthol AS-MX phosphate on peripheral blood from 34 normal, 31 pathological cases and several different species of animals with the following results.
1) Among the peripheral blood cells in normal subjects, alkaline phosphatase activity was confined to neutrophilic leukocytes only.
2) The neutrophils of the rat, rabbit, dog and sheep showed a varying degree of alkaline phosphatase activity.
3) Eight patients with aplastic anemia were examined and in all, high levels of activity were invariably found.
4) Five patients with chronic myelogenous leukemia were found to have a very low level of activity, one of them even being in full remission. The alteration in alkaline phosphatase activity during phases of changing leukocyte count in two patients treated with X-ray revealed that activity had beeb still below the normal range even after the full hematological and clinical remissions were induced.
5) The results of leukocyte alkaline phosphatase activity in various other hematologic disorders and also miscellaneous conditions were presented and theri significance discussed.
6) This As-MX method proved to be the most satisfactory as compared with the azo dye technic and Takamatsu-Gomori method because of it¢¥s higher sensitivity and better reproducibility.
